ABSTRACT

Since metabolic process differs between humans and mice, studies were performed in hamsters, which are generally considered to be a more appropriate animal model for studies of obesity-related metabolic disorders. The modulation of gut microbiota, bile acids and the farnesoid X receptor (FXR) axis is correlated with obesity-induced insulin resistance and hepatic steatosis in mice. However, the interactions among the gut microbiota, bile acids and FXR in metabolic disorders remained largely unexplored in hamsters. In the current study, hamsters fed a 60% high-fat diet (HFD) were administered vehicle or an antibiotic cocktail by gavage twice a week for four weeks. Antibiotic treatment alleviated HFD-induced glucose intolerance, hepatic steatosis and inflammation accompanied with decreased hepatic lipogenesis and elevated thermogenesis in subcutaneous white adipose tissue (sWAT). In the livers of antibiotic-treated hamsters, cytochrome P450 family 7 subfamily B member 1 (CYP7B1) in the alternative bile acid synthesis pathway was upregulated, contributing to a more hydrophilic bile acid profile with increased tauro--muricholic acid (TMCA). The intestinal FXR signaling was suppressed but remained unchanged in the liver. This study is of potential translational significance in determining the role of gut microbiota-mediated bile acid metabolism in modulating diet-induced glucose intolerance and hepatic steatosis in the hamster.

ABSTRACT

Objective To share for the nursing adverse events resources in the region, which would provide scientific data for nursing security management. Methods The form of nursing adverse events reporting was first designed by researchers. Then with the help of computer engineers the module on regional collaborative information platform was constructed. Preliminary running was used to modify and improve. Through continuous improvement the module function of adverse events reporting, query and statistics was finally made out. Results The module on regional collaborative information platform for nursing adverse events reporting and analysis was constructed, and it worked well. Conclusions The module realizes the sharing of nursing adverse events resources in the region. It can improve the level of nursing safety management in each hospital, which helps to reduce the incidence of nursing adverse events.

ABSTRACT

Objective To investigate the correlations of plasma homocysteine(Hcy)level and apolipoprotein E gene polymorphism with Alzheimer' s disease(AD)and mild cognitive impairment (MCI).Methods A case-control study in 66 AD patients(AD group),64 MCI patients(MCI group) and 54 healthy controls(control group)was conducted.Plasma Hcy level and ApoE polymorphism were determined and analyzed.Results Plasma Hcy levels were significantly higher in AD and MCI groups than in control subjects(both P<0.001).AD patients also showed increased plasma Hcy levels as compared with MCI patients(P<0.001).Logistic regression analysis indicated that the increased plasma Hcy level was a risk factor for AD and MCI(OR= 1.435 and 1.312,both P<0.001).ApoE ε3/3 was the most common genotype in AD,MCI and control groups,and ε3/4 and ε4/4 genotypes were more common in AD group and MCI group than in control group(both P<0.05).The ε4 allele frequency of ApoE was 24.2% and 23.4% in AD or MCI group respectively,and 6.5% in control group(AD or MCI vs.control,P<0.05).The analysis by multiplicative interaction model showed that the odd ratio for MCI was 23.3 in patients with only hyperhomocysteinemia(Hhcy,Hcy> 15 μmol/L),12.6 in patients with carrying ε4 allele,and 46.7 in patients with both Hhcy and carrying ε4 allele,which indicated that there was interaction between hyperhomocysteinemia and carrying e4 allele.Conclusions Hyperhomocysteinemia and ApoE ε4 allele are correlated with dementia and also have additive interactions.

ABSTRACT

Objective To build the dynamic management system of nursing staff allocation in general pediatric wards based on working hours measurement.Methods 30 general pediatric wards of 25 general or children hospitals of upper first class were selected to measure working hours.Depending on the average nursing hours required for pediatric patients in different care levels by each positions,we achieved the nursing staffing formulas.Results 7 nursing posts under double-shift turn and three-shift turn were collected.The average hour that nursing posts provide for one different nursing grade patient was made out.Taking a ward which had 20 patients as nursing grade one and 20 patients as nursing grade two for example,it would need 8-9 responsible nurses,2-3 long night shift nurses,1-2 general affairs nurses,2 night assistance nurse,1 day assistance nurse by double-shift turn,meanwhile,it would need 8 responsible nurses,4 early night shift nurses,3 late night shift nurses,1-2 general affairs nurses by three-shift turn.Conclusions Using the formulas based on measurement of working hours to calculate the number of nursing staff could provide a way to help the managers.

ABSTRACT

Objective To detect the treatment effect of PC3 cells with triptolide on altering the expression of genes.Methods MTT assay was used to detec the inhibition of proliferation.Apoptosis was detected by Annexin-V/PI staining.RT-PCR was used to analyze the mRNA expressions of BCL-2,BAX,PIG3,P21,FAS,CASPASE3.Results Triptolide caused a time - and dose - dependent inhibition of cell proliferation,and IC50 of 24 h and 48 h were 18.3 ng/ml and 13.5 ng/ml,respectively.Compared with the 24 h group,the low concentration of triptolide(5 ng/ml,t =1.47,P >0.05)and the high concentration of triptolide ( 160 ng/ml,t =0.91,P >0.05)had no statistical significance in 48 h group,while 10 ng/ml( t =3.26,P <0.05),20 ng/ml( t =4.21,P <0.05),40 ng/ml( t =4.09,P <0.05),80 ng/ml( t =2.91,P < 0.05 )had statistical significance.At the concentration of 18.3 ng/ml,triptolide induced PC3 cells apoptosis in a time - dependent manner.Compared with the control group,Anexin-V( + )/PI(-)was(5.42±2.21)%(t =3.52,P <0.05)in 6h,(13.51±3.37)%(t =6.53,P <0.01) 12h,(29.3 ±4.53)% ( t =8.74,P <0.01) 24 h group separately,and it had statistical significance.RTPCR showed that 18.3 ng/ml triptolide up-regulated the mRNA expression of BAX and PIG3,down-regulated P21 and BCL-2.FAS and CASPASE3 did not show obvious changes.Conclusions Triptolide inhibits the proliferation of PC3 and induces apoptosis,and the changes of BCL-2,BAX,PIG3 and P21 may play an important role in the apoptosis of PC-3 cells.
